چکیده

Using a rat model we have previously proposed a role for fibronectin as an adhesive ligand on high endothelial cells (HEC) for recirculating lymphocytes. Lymphocyte adhesion to high endothelial cells was blocked by CS1 peptide (from the type IH connecting segment of fibronectin) and RGD-containing peptides using two different in vitro assays of lymphocyte-HEC recognition, the frozen section assay and cultured HEC. In order to study the receptors utilised by lymphocytes to bind to HEC we have developed a xenogeneic model in which the adhesion of human lymphocytes to HEC cultured from rat lymph nodes is measured. The basic properties of lymphocyte-HEC interaction were retained using human lymphocytes. CS1 peptide and RGD-containing peptides gave similar profiles of inhibition of lymphocyte adhesion as found previously using rat cells.

Different Stages of Platelet Adhesion to the Site of Vascular Injury

Platelet activation and adhesion to the site of vascular injury is a dynamic process comprising reversible and irreversible phases. Platelet adhesion typically occurs in a multi-step process similar to the selectin/integrin-mediated adhesion of neutrophils. This phenomenon is highly regulated and influenced by the cross-talk between platelets and injured endothelium. This cross-talk involves a ...

CD44-mediated activation of α5β1-integrin, cortactin and paxillin signaling underpins adhesion of basal-like breast cancer cells to endothelium and Fibronectin-enriched matrices

CD44 expression is elevated in basal-like breast cancer (BLBC) tissue, and correlates with increased efficiency of distant metastasis in patients and experimental models. We sought to characterize mechanisms underpinning CD44-promoted adhesion of BLBC cells to vascular endothelial monolayers and extracellular matrix (ECM) substrates. Stimulation with hyaluronan (HA), the native ligand for CD44,...

The C–C Chemokine MCP-1 Differentially Modulates the Avidity of b1 and b2 Integrins on T Lymphocytes

cytes into inflammatory sites in vivo (Bargatze and The ability of chemokines, particularly MCP-1, to inButcher, 1993; Spangrude et al., 1985). This implicates duce integrin-dependent binding of T lymphocytes to a G protein–coupled signaling event in lymphocyte adendothelial adhesion molecules or extracellular matrix hesion and extravasation and suggests a role for chem(ECM) components was exam...

Chemokine stromal cell-derived factor-1alpha modulates VLA-4 integrin-mediated multiple myeloma cell adhesion to CS-1/fibronectin and VCAM-1.

The chemokine stromal cell-derived factor-1alpha (SDF-1alpha) and its G-protein-linked receptor CXCR4 are involved in hematopoietic progenitor cell and lymphocyte migration. The integrin VLA-4 is a cell adhesion receptor for CS-1/fibronectin and VCAM-1 and constitutes one of the main adhesion receptors mediating myeloma cell adhesion to bone marrow (BM) stroma in multiple myeloma (MM). It is sh...

Rab11 is required for maintenance of cell shape via βPS integrin mediated cell adhesion in Drosophila

In eukaryotes, vesicle trafficking is regulated by the small monomeric GTPases of the Rab protein family. Rab11, (a subfamily of the Ypt/Rab gene family) an evolutionarily conserved, ubiquitously expressed subfamily of small monomeric Rab GTPases, has been implicated in regulating vesicular trafficking through the recycling of endosomal compartment. In an earlier communication, we have shown th...
